Indications/Uses
Diphtheria, pharyngitis, rheumatic fever, spleen disorders & syphilis.
Dosage/Direction for Use
Upper resp infections Adult 1,200,000 u. Childn ≥27.3 kg body wt 900,000 u, ≤27.3 kg body wt & infant 300,000-600,000 u. Doses to be given as single IM doses w/ a frequency of 2 inj in 10 days. Syphilis 2,400,000 u as a single IM dose, to be given once a wk for 3 wk. Congenital syphilis Childn 2-12 yr ¼-1/3 the adult dose, ≤2 yr & infant 50,000 u/kg body wt as a single IM dose. To be taken once a wk for 2 wk. Prophylaxis of rheumatic fever & Strep pharyngitis 1,200,000 u as a single IM dose every 3 wk.
Contraindications
Hypersensitivity to penicillins. Do not inject into or near an artery or nerve.
Special Precautions
Hypersensitivity to cephalosporins & other β-lactam antibiotics, poor renal function. IV & intrathecal inj should be avoided. History of cardiac decompensation, severe heart disease.
Adverse Reactions
Skin rash, anaphylaxis, hemolytic anemia, angioedema, urticaria, acute interstitial nephritis.
Drug Interactions
Bacteriostatic antibacterial agents, probenecid.
