Indications/Uses
Tab: Treatment of infections where β-lactamase-producing organisms might occur including uncomplicated gonorrhea, otitis media, resp tract infections & UTI. Inj: Treatment of upper & lower resp tract & gynecological infections. Prophylaxis to reduce the risk of infections following surgery or caesarean section during birth.
Dosage/Direction for Use
Tab Adult 375-750 mg bid. Uncomplicated gonorrhea 2.25 g w/ probenecid 1 g. Inj Adult 1.5-3 g 6 hrly. Acute pelvic inflammatory disease 3 g 6 hrly in conjunction w/ doxycycline (100 mg 12 hrly by oral or IV). Parenteral regimen may be discontinued 24 hr after clinical improvement; oral doxycycline should be continued to complete the 14 days of therapy. Childn ≥1 yr 300 mg/kg daily by IV infusion in equally divided doses 6 hrly; ≥1 mth Mild to moderate infection 100-150 mg/kg daily in 4 divided doses.
Administration
May be taken with or without food.
Contraindications
Hypersensitivity to penicillins.
Special Precautions
Patients w/ mononucleosis. Overgrowth of nonsusceptible organisms & superinfections. Hepatic, renal & hematopoietic dysfunction. Pregnancy & lactation. Neonates & infants.
Adverse Reactions
Serious anaphylactoid reactions. Diarrhea or loose bowel, nausea, vomiting, epigastric distress & abdominal cramps; anemia, thrombocytopenia, eosinophilia & leukopenia; transient elevations of ALT (SGPT) & AST (SGOT) transaminases, bilirubinemia, abnormal hepatic function & jaundice; rash, itching & other skin reactions.
Drug Interactions
Increased risk of bleeding w/ warfarin & methotrexate. May decrease efficacy of estrogen-containing OCs. Reduced excretion w/ probenecid. Allopurinol, aminoglycosides.
