Description

Indications/Uses

Infections due to β-lactamase-producing strains (where amoxicillin alone is not appropriate) including resp tract, genitourinary & abdominal infections; cellulitis, animal bites, severe dental infection w/ spreading cellulitis.
 

Dosage/Direction for Use

Adult & childn >12 yr Mild to moderate infections 375 mg 8 hrly or 625 mg 12 hrly. Dental abscess 375 mg 8 hrly for 5 days. Severe infections including chronic & recurrent UTI & lower resp tract infections 625 mg 8 hrly or 1 g 12 hrly. Hemodialysis 375 or 625 mg 24 hrly. Renal impairment CrCl 10-30 mL/min 375 or 625 mg 12 hrly, CrCl <10 mL/min 375 mg 12 hrly.
 

Administration

May be taken with or without food: Best taken at the start of meals for better absorption & to reduce GI discomfort.
 

Contraindications

Hypersensitivity to penicillins. Cross-sensitivity w/ other β-lactam antibiotics eg, cephalosporins. Previous history of jaundice/liver impairment associated w/ penicillins. Glandular fever or lymphatic lymphoma.
 

Special Precautions

Hepatic & renal impairment. Pregnancy & lactation.
 

Adverse Reactions

Hypersensitivity reactions & GI disturbances. Reversible leukopenia & thrombocytopenia, thrombocytopenic purpura, agranulocytosis, anemia, slight thrombocytosis, eosinophilia, abnormal platelet aggregation, prolonged bleeding & prothrombin time. Dizziness, headache, reversible hyperactivity & convulsions, agitation, anxiety, behavioral changes, confusion, insomnia. Increased AST & ALT, serum bilirubin & alkaline phosphatase; hepatitis, cholestatic jaundice, acute hepatic dysfunction. Acute interstitial nephritis, crystalluria, vag itching, soreness & discharge. Superficial tooth discoloration.
 

Drug Interactions

Probenecid, allopurinol, methotrexate, OCs, warfarin, disulfiram.
