Indications / Uses

PEFTRIN Ceftriaxone 1g Powder for Injection is used for treating a wide range of severe bacterial infections. These infections include lower respiratory tract infections, skin and soft tissue infections, bone and joint infections, and complicated urinary tract infections, including pyelonephritis. It is also used in the management of bacterial meningitis, bacterial endocarditis, and for surgical site infection prophylaxis.

Side Effects

PEFTRIN Ceftriaxone 1g Powder for Injection common side effects include: rash, diarrhea, injection site reactions (pain, tenderness, induration), and abnormalities in blood cell counts such as eosinophilia, leukopenia, and thrombocytopenia. Less common effects may involve elevated liver enzymes, headache, dizziness, nausea, and vomiting. Severe reactions such as anaphylaxis, pseudomembranous colitis, and gallstone or kidney stone formation are rare but possible.

Directions and Dosage

  • Dosage for Adults and Children over 12 years (≥ 50 kg): The usual daily dose is 1 - 2 g administered once daily. For severe infections, the dose can be increased up to a maximum of 4 g once daily.
  • Dosage for Infants and Children 15 days to 12 years (< 50 kg): The general dosage is 50-80 mg/kg of body weight once daily, depending on the severity and type of infection. The maximum daily dose should not exceed 4 g. For most serious non-meningitis infections, doses of 50-75 mg/kg/day divided every 12 hours may be used.
  • Dosage for Neonates (birth to 14 days): 20-50 mg/kg body weight once daily. The maximum daily dose must not exceed 50 mg/kg.
  • For specific conditions (e.g., Bacterial Meningitis in children): 100 mg/kg/day (maximum 4 g) in a single daily dose or divided every 12 hours for 7-14 days.
  • For Uncomplicated Gonorrhea (Adults): A single intramuscular dose of 500 mg.
  • For Surgical Prophylaxis (Adults): 1 g administered as a single dose 0.5-2 hours before the procedure.

The duration of treatment varies depending on the type and severity of the infection. Dosage adjustments are typically not necessary for patients with kidney or liver impairment if the other organ function is normal.

Contraindications

This should not be used in individuals with a known allergy to the active ingredient, any other cephalosporin antibiotics, or other beta-lactam antibiotics like penicillin. It is also contraindicated in newborns with hyperbilirubinemia, especially premature infants. Should not be given concomitantly with calcium-containing intravenous solutions, including continuous calcium-containing infusions, in any age group due to the risk of potentially fatal precipitation.

Special Precautions

  • Requires special caution in people with a history of gastrointestinal diseases, particularly colitis.
  • Should be used with care in people with a combined impairment of both liver and kidney function.
  • Prolonged use may lead to overgrowth of non-susceptible organisms.
  • Risk of hemolytic anemia, which can be severe, should be monitored.

Is it safe to take it with other medications?

Should not be mixed or administered simultaneously with calcium-containing solutions via the same IV line. Combining it with oral anticoagulants may increase the risk of bleeding. The concurrent use of other aminoglycoside antibiotics should be carefully considered due to potential increased risk of kidney toxicity.

How should I store it?

Store the powder for injection below 30°C. Protect it from light. The reconstituted solutions should be used immediately or stored briefly as instructed on the product leaflet.
